Pre-Doctoral and Post-Doctoral Fellowship Grant

ChadTough Defeat DIPG Foundation

The ChadTough Defeat DIPG Fellowship Grant is intended to attract and support promising scientists embarking on careers involving DIPG research. By providing funding to outstanding fellows under the guidance of a mentor, we seek to assist in the development of the next generation of leaders in the field of DIPG research. The ChadTough Defeat DIPG Fellowship Grant is available to pre-doctoral and post-doctoral researchers. The amount of the grant is up to $200,000 over 3 years (maximum of $67,000 per year) for pre-doctoral fellows, and up to $300,000 over 3 years (maximum of $100,000 per year) for post-doctoral fellowsarch 1, 2030

Review and Selection Process

● Each application is reviewed by the ChadTough Defeat DIPG Scientific Advisory Council.

● The Scientific Advisory Council makes recommendations to the ChadTough Defeat DIPG Foundation, and the foundation makes the final funding decision.

● All application evaluations are considered confidential and are available only to the Scientific Advisory Council and the foundation’s Board of Directors and staff.

● When an application is approved for funding, the grant award is contingent upon all legal documents being signed and approved by the foundation and the applicant’s institution.

Conditions of Grants

 We fund research to better understand the causes of DIPG and to advance its treatment and cure. To help achieve this goal, we expect researchers to publish their findings in peer-reviewed journals and to make their data available to other researchers.

 All grants are payable to the grant recipient’s institution or laboratory.

 The post-doctoral grant is to cover salary and fringe benefits only. The pre-doctoral grant is a stipend to cover living expenses and for the cost of tuition and fees. No funds may be used for indirect costs.

 Grant is limited to covering the applicant’s research time, not clinical time.

 Overlap in funding is not permitted. This includes scientific, budget, and commitment overlap, as defined by the National Institute of Health.

 Requests for transfers to another institution while the grant is in effect will be considered on a case-by-case basis.

 No-Cost Extensions may be requested by emailing grants@chadtough.org.

 Grant recipients will work with us to coordinate a joint press release to announce the grant.

AI Based Application Success Predictor

🧠 1. Strict Focus on DIPG/DMG (Non-Negotiable)

  • Research must directly address:
    • Diffuse Intrinsic Pontine Glioma (DIPG) or Diffuse Midline Glioma (DMG)
  • These are rare, aggressive pediatric brain tumors with extremely poor prognosis

👉 Non-DIPG brain tumor work → very low success probability

🚀 2. Breakthrough-Level Innovation (Top Predictor)

  • Core mission: fund “novel approaches” and “game-changing research”
  • Highest-funded category:
    • Game Changer Grants → explicitly for bold, transformative ideas

👉 Incremental research is rarely competitive

💊 3. Clear Path to New Therapies (Critical)

Successful proposals typically:

  • Aim to:
    • Develop new drugs or treatment strategies
    • Target key mutations (e.g., H3K27M)
  • Example funded work:
    • First-in-class drug candidates targeting DIPG mutations

👉 Strong emphasis on therapeutic development

🔄 4. Translational & Clinical Trajectory

  • Projects should move toward:
    • Preclinical validation → clinical trials
  • Many funded studies:
    • Focus on drug delivery, immunotherapy, or targeted therapy

👉 Pure basic science must show clear clinical pathway

🧬 5. Addressing Major Biological Barriers in DIPG

High-success proposals often tackle:

  • Blood–brain barrier (BBB) challenges
  • Tumor microenvironment
  • Genetic drivers of DIPG

👉 Example: innovative drug delivery systems to bypass BBB

⚡ 6. High-Risk, High-Reward Approach

  • Foundation explicitly encourages:
    • Bold, unconventional ideas
  • Especially:
    • “Undruggable” targets
    • New therapeutic platforms

👉 Risk is acceptable if potential impact is huge

🤝 7. Collaboration & Global Research Integration

  • Strong preference for:
    • Multi-institution collaborations
    • Participation in global DIPG research ecosystem

👉 Foundation actively brings together “brightest minds” to accelerate progress

👩‍🔬 8. Investigator Profile (Two Pathways)

Early-career:

  • Fellowship grants for:
    • Pre/postdoctoral researchers
    • Strong mentorship required

New investigators:

  • Support for:
    • New labs OR researchers entering DIPG field

👉 Unique feature: encourages new entrants into DIPG research

🧪 9. Hypothesis-Driven, Mechanistic Science

  • Especially for Game Changer grants:
    • Must be hypothesis-driven research
    • Address a major unresolved challenge

👉 Strong mechanistic rationale is essential

📊 10. Potential for Field-Wide Impact

  • Projects should:
    • Advance overall understanding of DIPG
    • Generate broadly useful insights or technologies

👉 Foundation funds “most promising discoveries” globally

📊 Summary: Key Predictors (Ranked)

Highest impact factors:

  1. DIPG-specific focus
  2. Breakthrough innovation
  3. Clear therapeutic pathway

Moderate predictors:
4. Translational trajectory
5. Tackling major biological challenges (BBB, mutations)
6. High-risk/high-reward approach

Supporting factors:
7. Collaboration
8. Investigator potential
9. Mechanistic rigor

💡 Insider Insight

Compared to other pediatric cancer funders:

  • ChadTough Defeat DIPG Foundation is:
    👉 extremely innovation-driven + cure-focused

👉 Key evaluation question:
“Is this a bold idea that could realistically lead to a cure for DIPG?”

  • First-in-class therapy potential ✔️
  • Strong biology + translational link ✔️
  • High impact on a major barrier ✔️
    Highest success probability
